09-P002 The transcription co-factor Cited2 functions during sex determination and early gonad development

چکیده
The apical ectodermal ridge (AER), a thickening of the limb epithelium at its distal tip, is pivotal for maintaining vertebrate limb outgrowth. In a recent study, Flrt3 has been involved in the activation of ERK by FGFs. Moreover, Flrt3 has been described as a transmembrane protein able to interact with FGF receptors modulating, this way, FGFR signaling, either directly or recruiting cytoplasmic cofactors that enhance the activation of this pathway. Preliminary results show that Flrt3 expression is restricted to the AER, colocalizing its expression with fgf8 and pERK activity. Moreover, our data indicate that Flrt3 expression is not regulated by FGF activity, although ectopic Wnt3a is able to induce Flrt3 expression. Loss-of-function studies have shown that the silencing of Flrt3 affects the integrity of AER and, subsequently, its proper activity during limb bud outgrowth. We are performing studies in order to uncover the downstream targets o the Flrt3/Erk activity.

The transcription co-factor CITED2 functions during sex determination and early gonad development.
The early bi-potential mammalian gonad requires the expression of a Y-linked gene, Sry, during a brief window of time to ensure proper testis development. WT1 and its direct target gene Sf1 function during sex determination as well as in the specified testes and ovaries. We have previously shown that the transcription co-factor CITED2 interacts with WT1 to stimulate the expression of Sf1 in the...

It has been proposed that the mammalian adrenal cortex and gonad are derived from the same primordium present during early urogenital development. Molecular pathways involved in the differentiation of the adrenal cortex from the adrenogonadal primordium (AGP) have yet to be determined. Here we show in mice that the transcription co-factor Cited2 is required for the specification of the adrenal ...
متن کاملI-17: The Mechanism of Gonadal Sex Determination
Background In mammals, a single exon gene SRY on the Y-chromosome is activated in the XY gonadal primordium and initiates a cascade of molecular and morphological events leading to testicular differentiation. SRY-encoded protein (SRY) is a transcription factor harboring a HMG-box DNAbinding motif that upregulates SOX9, which encodes another transcription factor sharing the DNA binding motif wit...
متن کاملI-43: Identification of SOX3 as an XX MaleSex Reversal Gene in Mice and Jumans
Background: Mammals utilise an XX/XY system of sex determination in which the Y-linked gene SRY (Sexdetermining region Y) exerts a dominant masculinising influence on sexual development. Sex chromosome homology and comparative sequence studies suggest that SRY evolved from the related SOX3 gene on the X chromosome, although there is no direct functional evidence to support this hypothesis. The ...

In birds, the mechanism of sex determination is unknown: it is uncertain whether the female-specific W-chromosome encodes an ovary-determining gene analogous to the mammalian testisdetermining gene, or whether testis determination is dependent on a double dose of a Z-chromosome gene(s) that escapes inactivation. In addition, birds do not carry a homologue to the Sry gene and the identity of the...
